Adapting fish defenses to block human infections: Antimicrobial peptide of fish gills inspire clean surfaces

ScienceDaily (Nov. 29, 2012) ? An undergraduate research team analyzes peptides from fish gills to engineer antimicrobial surfaces for food preparation and medical devices.

Living in an environment teaming with bacteria and fungi, fish have evolved powerful defenses against waterborne pathogens, including antimicrobial peptides located in their gills. Undergraduate researchers at Worcester Polytechnic Institute (WPI) are studying the biology and the mechanics of one of those peptides with the hope they can use that knowledge to create engineered surfaces that kill bacteria responsible for foodborne illnesses and hospital-acquired infections.

The research team, led by Terri Camesano, professor of chemical engineering, reports its latest findings in the paper "Creating Antibacterial Surfaces with the Peptide Chrysophsin-1," published online in October by the journal ACS Applied Materials & Interfaces.

"Fish have a wonderful solution for blocking bacterial and fungal infections," Camesano said. "In this study, we are working to better understand the biochemical mechanics of that process."

As fish filter water through their gills to extract oxygen, antimicrobial peptides (AMPs), including Chrysosphin-1, trap and kill pathogens before they can invade the fish's bloodstream. Scientists in many laboratories around the world are actively exploring the potential use of these molecules to prevent human infections. In the current study, the WPI team attached AMPs to silicon and gold surfaces using two different approaches and measured the bound peptides' ability to kill the bacterial pathogen E. coli.

In the first method, the AMPs were absorbed directly onto gold and silicon crystals, forming a single layer of molecules with the AMPs lying flat on the surface. In the second method, the tips of the AMPs were attached to the surfaces with a glue-like substance so that the peptides rose vertically, like blades of grass extending up from the ground. Surfaces with both AMP configurations were cultured with E. coli cells. The results showed that when the AMPs were lying flat they killed 34 percent of the bacteria in the culture, but when they were standing up vertically they killed 82 percent.

"The hypothesis is that when peptides are attached vertically to the surfaces, they are better able to move and bend so they take on a shape that is more effective in binding to and disrupting the E.coli cells," Camesano said.

In addition to gathering data about the antibacterial efficacy of the attached AMPs, the WPI research team developed a technique for monitoring, in real time, the attachment of AMPs to surfaces. Using quartz crystal microbalance with dissipation monitoring (QCM-D), the team measured the quantity of AMPs that successfully attached to the surfaces in the horizontal and vertical orientations and the density of the AMP layers, along with other properties.

"This was a powerful process, to be able to essentially watch the binding process as it happened," Camesano said. "It is a technique that we will continue to apply in further studies."

Camesano said gold and silicon surfaces were selected for the current study because their chemical properties are well-suited for AMP binding. In ongoing work, Camesano's laboratory will continue to characterize the mechanics of AMP binding for optimal antimicrobial activity and test other materials, including titanium, stainless steel, and plastics, that would have greater utility in food preparation and healthcare.

"What is also notable about this study is that it is the work of undergraduates," Camesano said. "They've done excellent work here that will inform future graduate studies in our lab."

The WPI undergraduates who co-authored the AMP paper are Ivan Ivanov '12 (chemical engineering), Alec Morrison '12 (biochemistry), and Jesse Cobb '12 (chemical engineering). Co-author Catherine Fahey, a student at George Washington University, worked on the study during at a summer project at WPI. Their work was sponsored in large part by a grant from the National Science Foundation's Research Experiences for Undergraduates (REU) program.

Sony Xperia Tablet S sales resume

Sony has resumed sales of its Xperia Tablet S one month after pulling it from the market.

The company withdrew the slate in early October after discovering a build defect that allowed moisture to seep in between its screen and casing.

Sony previously confirmed on its Japanese website that the product would gradually return to stores in November, a promise it kept with just days to spare.

Sony had already distributed around 100,000 Xperia S tablets in Europe, the US and Japan when the flaw was detected, but honoured a free inspection and repairs agreement for users affected by the issue.

Running Android 4.0 Ice Cream Sandwich, the tablet is powered by a speedy NVIDIA Tegra 3 quad-core processor and is backed by an extensive media library courtesy of the Sony Entertainment Network.

The Sony Xperia Tablet S retailed in the UK from ?329, with the price tag climbing for models with higher memory.

How does a volcanic crater grow? Grab some TNT and find out

ScienceDaily (Nov. 26, 2012) ? A new University at Buffalo study in the journal Geophysical Research Letters examines maar craters, which resemble the bowl-like cavities formed by meteorites but are in some ways more mysterious.

Scientists often can discern pertinent details about meteorites -- when they struck, how large they were, the angle they approached Earth and other information -- by measuring the diameter and volume of the impact crater.

Maar craters, which form when fissures of magma beneath Earth's surface meet groundwater, causing volcanic explosions, are not as telling, scientists say. The possibility of multiple explosions at varying depths led most scientists to believe that measuring a maar's size is not the best way to gauge the energy of individual explosions or determine future hazards.

UB geologist Greg A. Valentine, PhD, and other volcano researchers found instead that examining a maar's shape and the distance it ejects magma, ash and other debris to be a more accurate barometer of the eruption's force. The findings are important, he said, because they could assist scientists in estimating how big future volcano eruptions might be.

"It's something that, up until this point, had only been suspected," said Valentine, a professor of geology and lead author of the Geophysical Research Letters paper. "The simulations we did prove that crater diameter is not a good indicator of explosion energy for these volcanoes."

The scientists drew their conclusions on a series of UB-funded experiments conducted last summer at a test site in Ashford, N.Y. They built three test beds of gravel, limestone and asphalt. In the first experiment (see the video below) one charge of TNT and plastic explosive was detonated.

In subsequent experiments, the charge was divided into three parts and detonated individually at different depths. The final dimensions of each crater were about the same. That matters, according to Valentine, because it shows that it's easy to overestimate the energy of explosions if one assumes that the crater comes from one blast, not several.

The dispersal of ejected material differed depending on the location of the charge. For example, the first experiment launched debris more than 50 feet from the crater. Debris from subsequent experiments simulating blasts further underground mostly went up in the air and fell back into the crater or around its rim. As a result, it forced dusty gas -- like the ash that shut down air travel in Iceland and beyond in 2010 -- into the surrounding air. This can be seen in the video below.

Although the experiments provided valuable information, Valentine said they were similar to a practice run. More detailed experiments are being planned for the near future, he said.

These types of seal are, however, only best suited to the most narrow window gaps, so won?t always be the best option for older, larger properties or sash windows.
  • Triple Glazing ? if you already have double glazing, then compression seals aren?t usually recommended ? otherwise you allow almost no room for natural ventilation, which is important in preventing the build-up of mould and onset of rising damp in a household room. What you can still do, however, is have triple glazing installed onto the existing window; that way you improve noise reduction levels and heat insulation without running the risk of affecting you and your family?s health.
  • Air-Vacuumed Windows ? double and triple glazing also often comes with the optional service of air-vacuuming; that is, a contractor injects inert gas (often argon or something similar) in the gaps between each glass pane. This improves the insulating properties of the window and, in a similar fashion as the glazing itself, slows down any potential ?draughts? keen to enter your home.
  • Window Draught Excluder ? if you?re looking for something a little cheaper than draught seals or extra glazing you can also find a range of ready-to-use window draught excluders from a range of high-street stores, hardware outlets and online retailers. These aren?t necessarily the most effective way of dealing with draughts, and can only really be used at the bottom-most level of the pane ? so they don?t solve the entire problem either. They do, however, often come in a range of novelty designs and are a fun way of brightening up a room without having to worry about any serious DIY work.

So do you need to know how to draught proof windows in your home?

Well, if you are proficient in DIY and fancy taking on the job yourself, then there is no reason why you shouldn?t get to work as soon as your plan and budget are sorted! You might even want to consider installing extra measures, such as draught proofing your doors to ensure you home is as warm and energy efficient as possible.

If you lack the confidence to do it yourself, however, it is strongly recommended you hire a professional draught proofing contractor to install the seals for you. That way you know your home is draught proofed properly and you don?t have to worry about wasting the time and money on giving it a go yourself. Plus a fully qualified surveyor can carry out a Fan Pressurisation Testbefore work begins, so all the draughts are expertly located and you don?t waste money on any seals where they?re not needed! Just remember to compare a number of different quotes and make sure your contractor is affiliated with British Standards BS7386 for draught-proof guidelines before you make any final decisions on who to hire.

Gaza cleric calls violation of Israel truce sinful

GAZA CITY, Gaza Strip (AP) ? A leading Islamic cleric in the Gaza Strip has ruled it a sin to violate the recent cease-fire between Israel and the Hamas militant group that controls the Palestinian territory.

The fatwa, or religious edict, issued by Suleiman al-Daya late Saturday accords a religious legitimacy to the truce and could justify any act by Gaza's government to enforce it.

"Honoring the truce, which was sponsored by our Egyptian brethren, is the duty of each and every one of us. Violating it shall constitute a sin," the fatwa read.

The Wednesday truce put an end to an eight-day Israeli offensive against Gaza militants who fired rockets into Israel. The agreement remains fragile because details beyond the initial cease-fire have not yet been worked out.

Russia's MegaFon gets nod for $2 billion IPO - sources

MOSCOW (Reuters) - Russia's No.2 mobile phone company, MegaFon , is likely to go ahead with its planned stock market float in London this year after receiving an unofficial green light from the UK regulator, sources familiar with the matter said.?

MegaFon had aimed to raise around $2 billion (1 billion pounds) from its initial public offering this year, but several sources have said it would have to delay the share sale until 2013 if it did not get the nod from the UK Listing Authority (UKLA) in time.

MegaFon has cleared and resolved any material issues with the UKLA, a source familiar with the situation said on Saturday. Official approval is expected when MegaFon's IPO prospectus has been completed, the source said.

?The company is expected to make a formal statement on the IPO launch in the coming weeks. The deal could happen in late November or early December, before capital markets wind down for the end of the year, sources have said.

The float by MegaFon, which is controlled by Russia's richest man Alisher Usmanov, would rank as the biggest by a Russian company since Internet search firm Yandex raised $1.4 billion when it floated in New York in May 2011.

LISTING CRITERIA

The UKLA approves an IPO prospectus when it is satisfied a company has met its listing criteria. Among issues the UKLA typically examines is the shareholder structure. Any particular concerns are typically listed in the 'risks' section of the prospectus.

Usmanov told Reuters in September that he would fold his stake in MegaFon and other assets into a new holding company, co-owned by one of his partners Vladimir Skoch, father of billionaire Russian lawmaker Andrei Skoch, who helped build up iron ore miner Metalloinvest.?

?Goldman Sachs dropped out as an underwriter of the IPO in October, which one source familiar with the situation said at the time was over unspecified shareholding concerns that were not related to Usmanov.?

?The UKLA was now satisfied with the information provided with regard to the new holding company structure, one of the sources said.

?The UKLA gave the go-ahead for the offering on Friday, said a second source familiar with the situation, meaning the float can happen this year. A third market source also said the IPO had received the green light but gave no more details.?

?A fourth source familiar with the matter said work still remained to be done on the prospectus, indicating a final sign off may not yet have been received.

?"We intend to complete the IPO this year and that will be on the basis of an approved prospectus from the UKLA," said a spokesman for MegaFon. He declined to comment further.?

?A spokesman for the UKLA declined comment.?

MegaFon announced plans for the offering in early October but later said it would put off launching the roadshow until it had published its third-quarter results.

Those results, released on Thursday showed gains in revenues, earnings and margins that analysts said would boost MegaFon's goal of achieving a valuation comparable with its competitors, MTS and VimpelCom .

Sources have said a stake of around 15 percent in MegaFon could be offered to investors in London and Moscow, with Nordic telecoms firm Teliasonera selling down its stake.

MegaFon also plans to pay down some of the debts it took in on in a restructuring deal in April that saw tycoon Mikhail Fridman exit the business and Usmanov secure overall control.

Known as the Opera Web Pass, the service is geared to those without data plans and allows users to purchase short term access from their local carrier. In its current form, mobile providers are given the flexibility to determine which subscriptions to offer, such as an hour or day of internet use, or even quick access to individual apps like Facebook or Google+. Naturally, the service could also be a great value for travelers. For its part, Opera insists that Web Pass should be extremely easy for carriers to implement with their servers. For the time being, however, Opera Web Pass is currently only available to subscribers of DiGi Telecommunications, which partnered with the Norwegian company to develop and test its service. Obama Bobblehead Launched Into Stratosphere (Romney, Too)

The U.S. presidential campaign was a political rollercoaster ride for Barack Obama and Mitt Romney, but it couldn't compare to the ride their bobblehead dolls took earlier this week.

On Monday (Nov. 5) ? the day before?President Obama defeated Romney?to win a second term in the White House ? a group of California schoolkids launched bobbleheads of the two candidates to the stratosphere aboard a high-altitude balloon.

"We estimate that the balloon reached 120,000 [feet]," Earth to Sky Calculus, a group of middle- and high-schoolers from Bishop, Calif., wrote on Facebook Wednesday. "It was a crystal clear, gorgeous fall day in the western United States!"

Onboard cameras documented the flight, recording the dolls' rise into the skies over east-central California and their dizzying descent after the balloon popped high above the Earth.

One video of the Obama and Romney dolls' flight, in fact, captures the transition from serenity to chaos after the balloon pops. The Obama doll's head is peacefully surveying the desert-mountain landscape one moment and bobbling furiously the next.

Earth to Sky Calculus eventually tracked down the fallen balloon and recovered its payload, group members said on Facebook.

Monday's launch wasn't the first?balloon flight?for some of the Bishop schoolkids. They also lofted a balloon in early September, as part of a project called The Golden iPod.

The Golden iPod is a nod to the Golden Records aboard NASA's?Voyager spacecraft, which launched in 1977 and are now nearing the edge of interstellar space. The records are meant to provide any aliens who may come across them an introduction to our species and our planet; they contain selections of some of our most celebrated music, greetings in 55 different human languages and much more.

The Golden iPod project aims to fill a 16-gigabyte mp3 player with more indicators and accomplishments of human culture, then launch it to Earth orbit in 2013.

"We plan to fill our iPod with the best humanity has to offer ? old and new," team member Anna Herbst said in a statement. "Then, if all goes as planned, we're going to send it to space."

Before it leaves the planet, the Golden iPod will visit the stratosphere during a series of high-altitude balloon flights, team members said. The first flight took place Sept. 5.

Trade Unions Attempt To Boost Maori Employment | Stuff.co.nz

Trade unions are trying to forge closer relationships with iwi groups to boost employment for Maori, but the Government is still being blamed for the latest rise in joblessness.

The number of unemployed Maori rose from 12.8 per cent in June to 15.1 per cent in the three months to September. Comparatively unemployment among New Zealanders of European ethnicity grew just 0.2 percentage points to 5.4 per cent from the previous quarter, while the overall national unemployment rate went up 0.5 points to 7.3 per cent.

Council of Trade Unions vice-president Syd Keepa said he had seen many more redundancies since the end of the September quarter and he would not be surprised to see the rate go higher.

He said Maori were traditionally employed by manufacturers such as sawmills which were proving uncompetitive against Asian manufacturers, especially with the high New Zealand dollar.

"I don't know how much of an effect the building programme in Canterbury's going to have . . . but in terms of building [products], that's where we're seeing a lot of our redundancies in manufacturing basically," Keepa said.

He's unhappy with the Government's decision to cut staff in the public service and the possible closure of the Hillside Engineering Workshops after train manufacturing contracts were awarded to overseas companies.

He said many Maori had moved to Australia to find paid work and believed the unemployment figure would be "horrendous" if that option was not open to them.

"I see some training being done but unfortunately a lot of those trained people go to Australia . . . it's good to get people trained up but where are the jobs? That's the reality, there are no jobs."

In September the CTU opened discussions with iwi leaders at a hui in Tauranga with a view to creating jobs for Maori, he said.

No concrete solutions had been reached yet.
